Personally i attacked Mamluks (before they lost any major war against Ottomans, i may add) directly in my recent ethiopian campaign after getting event with gifts from Portugal. I won.
So to be fair by that point i had much bigger territory under my control. About Defender of Faith problem - only Afrikan muslim states are under protection of Mamluks as DoF. Arabian states have no protection. So attacking arabian allies of protected states to get required territory is viable option.
