Potato tuber formation in tissue culture generally occurs around 4-6 months based on our experience. The use of tissue culture has been proposed and demonstrated as a clean mass production system by many publications (Akita and Takayama 1994; McCown and Joyce 1991). This particular culture is a Red potato variety after 6 months on just an MS-based medium with only 3% sucrose and 0.8% agar.

Flowering is a rare event in tissue culture during multiplication (Stage 2). Probably 1 in 500 plantlets. Shown here is Dianthus Arctic fire. Our QC group works hard to put every lot of media and components in test on various species, ensuring that each lot is viable and meets our quality standards. So it is a treat when we see a plantlet with vibrant flowers like this!

This particular culture was grown on our MS Basal Medium with Gamborg Vitamins (M404: https://hubs.ly/Q01c25-N0), supplemented with NAA and Kinetin.

Cellulases were first used to form plant-cell protoplasts 60 years ago (Cocking 1960). One of the earliest applications was somatic hybridization through protoplast fusion, used to create novel plant varieties (Evans 1983). Cellulases have also been used to form protoplasts for genome editing through CRISPR (Shan et al. 2013), with well-established methods for certain genera, like Arabidopsis (Yoo et al. 2007). These cellulases are often used in combination with macerozyme to dissolve cell walls.

Both Cellulase Onozuka RS and Cellulase Onozuka R-10 are derived from Trichoderma viride. They are multi-enzymatic systems consisting of cellulase, α-amylase, hemicellulase, pectinase, and protease activity.

Macerozyme R-10 is derived from Rhizopus sp. It is a multi-enzymatic system consisting of high pectinase, high hemicellulase, and low cellulase activities.

Pectolyase Y-23 is derived from Aspergillus japonicas. It is a purified maceration enzyme containing two types of pectinase, endo-polygalacturonase and endo-pectin lyase, that will macerate plant tissue when stimulated. Additionally, Pectolyase Y-23 can isolate biologically active protoplasts when used in combination with cellulase.
