"Sitting Bull's great-grandson identified through DNA fragments"
[Note: They don't really detail what techniques were introduced and utilized, and this is from about a year ago, but still interesting.]
Scientists confirmed 73-year old Ernie LaPointe as the closest living descendant of the famed Lakota leader Sitting Bull by analyzing fragments of the historic figure's DNA.
Eske Willerslev, a professor of ecology and evolution at the University of Cambridge, normally focuses on ancient DNA to understand human genetic diversity and how different groups of people around the world are similar and distinct. But he was incredibly excited for the opportunity to study Sitting Bull's DNA.
Willerslev heard over a decade ago about LaPointe's efforts to claim Sitting Bull's remains for reburial, and Willerslev felt compelled to assist as an admirer of the iconic Native American Chief.
"I reached out because I'm an ancient DNA researcher. I told LaPointe, 'If you want to do this, I think I can help you.'"
Obtaining enough usable fragments of Sitting Bull's DNA from the small hair sample proved challenging because the hair was badly degraded after being stored at room temperature at the National Museum of Natural History for more than a century.
"There was very little DNA in the hair — way too little for established methods of DNA analysis," he said. "So we had to develop a new method."
It took researchers 14 years to find a usable bit of DNA. Researchers hope the new DNA technique will open more doors to testing ancient DNA.
“In principle, you could investigate whoever you want — from outlaws like Jesse James to the Russian tsar’s family, the Romanovs. If there is access to old DNA — typically extracted from bones, hair or teeth, they can be examined in the same way,” Willerslev said in the press release.

Writing A Family History
A lot of people think that genealogy is just a dry list of names and dates. But, with the simple addition of personal memories and family stories, genealogy can be much more than that.
Buried in the memories of your relatives are stories just waiting to be told. Even in your own mind, there are memories worth preserving. A recorded interview is an excellent way to preserve these stories and memories. A recorded interview has the additional benefit of preserving the person's voice for prosperity. (But remember cassette tapes are perishable and will eventually fade! So make sure you have a transcript of your interview and digital record the interview if possible.)
Here are a few tips for doing a recorded interview.
First, sit where there will not be a lot of background noise that will be picked up on the recording.
Second, make sure that your recording device is in good working order. If you are using batteries in a tape recorder, make sure that you have fresh batteries.
Finally, some people are intimidated with the idea of speaking into a recorder, so attempt to place it a little off to the side so that it is not staring them in the face while they are talking.
Remember, every family has skeletons in the closet, and some relatives may not want to talk about those things or have them generally known. It is possible for those facts to be withheld from the copies of the family history that are circulated to others for a specified period time or until after a certain event (like the death of someone, or the death of everyone in a certain generation). But by writing down the story now you can be certain that it will not be lost for all time, after all none of us can be assured of tomorrow.
The November 1973 issue of The New Era magazine carried an article writen by William G. Hartley entitled "Suggested Family Oral History Topics" on page 21. The article includes an excellent outline that you can use to help you organize yourself. It is available online by going to: https://www.lds.org/new-era/1973/11/suggested-family-oral-history-topics
You may find using an outline the easiest way to write a biography or do an oral history interview. While the list may seem long when you first look it, you will find that the general topics will help the person remember many stories. This outline is just a general guide to help you develop specific questions for the person you are interviewing.
Remember you want details, as many and as specific as possible. When talking about their childhood home, encourage them to remember details of what it was like: What type of house was it? How many bedrooms? Did they share a bedroom or even a bed with others? What type of yard did they have? What type of neighborhood was it? What types of appliances where in their house? You are a reporter digging for facts, follow your instincts and satisfy your curiosity when asking questions.
Even if you fear that you will not have enough time to cover everything, don't put off asking questions. First, every little bit that you can record now is something that will not be lost. And second, several short interviews are better than one marathon interview anyway.
The following is a list of questions that you can use to write a biography and/or use in an oral history interview. Not all questions will apply to every person. The questions concern personal history and historical events which may have happened during someone's lifetime.
Do your best to get complete, honest answers, but if someone feels a question is too personal or "prying" please feel free to skip that question. Remember if they feel threatened they may clam up about everything.
While I am sure everyone knows this, I will repeat it anyway... When someone has been kind enough to share information with you remember to tell them "Thank you!"

All About DNA!
WELCOME! This site is intended to be a comprehensive ancestral DNA clearinghouse for genetic genealogists, those new to ancestral DNA, and anybody else who would like to learn more about how genetic genealogy works.
Our main goal is to PUBLICLY spread awareness of the positive aspects of DNA/genetic genealogy and to provide information to the public in the hopes that more people will become interested in ancestral DNA.
Another priority for this site is to provide a starting place for those who have just begun their DNA journeys and need more information on what comes next.
Additionally, we hope to provide a well-resourced site for people who are at any stage of their DNA research. In some instances, we will refer you to other sites who already have great resources in place.
Lastly, we hope to educate and encourage others who may be considering the purchase of a DNA kit.
