The high-purity chondroitinase is mainly used for the determination of chondroitin sulfate (enzymatic hydrolysis
liquid phase method).
Chondroitin sulfate degrades chondroitin sulfate A, chondroitin sulfate B (dermatan sulfate), and chondroitin
sulfate C. This enzyme acts on and cleaves the β1-4 glycosidic bond between the disaccharide repeating units
through a β-elimination mechanism. The degradation products are mainly unsaturated disaccharides, which can
be detected by a UV photometer (232 nm).

Prothrombin（Factor II)
Bovine prothrombin is prepared from freshly collected bovine plasma and consists of single-chain polypeptides. The prothrombin complex (FXa, FVa, PL, and Ca++) activates prothrombin to produce the serine protease thrombin. The purity of prothrombin was determined by SDS-PAGE, and the purity of prothrombin did not decrease after 2-mercaptoethanol incubation.

anticoagulation, why monitor anti-Xa?
Currently, activated partial thromboplastin time (APTT) is a standard indicator for monitoring unfractionated heparin in clinical practice. Still, its detection results are affected by many factors. Especially when there are lupus anticoagulants, coagulation factor inhibitors, and coagulation factor deficiencies in the patient's body, the APTT is usually prolonged. Some common external biological factors may also affect the test results. For example, during women's menstrual cycle, the APTT test may erroneously increase or decrease in the morning and evening tests, and the consequences for elderly patients are higher.
Further correction experiments are needed to determine the cause. That is, common causes of APTT prolongation include lupus anticoagulant (L.A.C.), congenital (hemophilia) or acquired (liver disease, warfarin, D.I.C.) factor deficiencies, etc. In contrast, common causes of APTT shortening include inflammation (leading to high levels of VIII factors, fibrinogen), pregnancy, kidney disease, etc. In addition, APTT prolongation is not evident with low molecular weight heparin therapy (LMWH), so anti-Xa (Anti-Xa) should be considered in these settings to monitor clinical efficacy. The monitoring of anti-Xa factor activity has a better inter-laboratory correlation than APTT. The detection results can reliably reflect the content of unfractionated heparin or low molecular weight heparin in the body.

The new coronavirus still needs to enter human cells

In a recent study, researchers from the University of California, San Diego School of Medicine found that SARS-CoV-2 could not catch ACE2 without a carbohydrate called heparan sulfate. Heparan sulfate on the lung's surface cells acts as the virus's co-receptor to enter cells.

Two methods can reduce the ability of SARS-CoV-2 to infect human cells cultured in the laboratory by about 80% to 90%. First, use enzymes to remove heparan sulfate. Second, use heparin as bait to lure and combine this coronavirus to keep it away from human cells. As a form of heparan sulfate, heparin is a widely used drug to prevent and treat blood clots, which indicates that a drug approved by the U.S. F.D.A. may be reused to reduce viral infections.

Esko said these findings are far from being translated into treatments for COVID-19 patients. People will need to test heparin and heparan sulfate inhibitors in animal models of SARS-CoV-2 infection. In a related study, scientists at the University of California, San Diego, are still exploring the human microbiome, including bacteria that live inside and on the body, influencing heparan sulfate to affect human COVID-19.
