Some of the most recent variants of concern and interest include Delta, Mu and Omicron.
Detection and analysis of viruses via Polymerase Chain Reaction (PCR) based methods or other techniques rely on molecular properties that could get affected by the mutation so it is important to monitor variants from a diagnostic perspective. For example, PCR methods rely on replication and detection of the genetic material (ribonucleic acid or RNA) of SARS-CoV-2 so if a mutation alters the RNA of the virus, the chemical replication PCR process may be hampered and the virus undetected. This is typically de-risked by amplifying multiple RNA regions.

Alternative detections methods, such as the SARS-CoV-2 LC-MS Kit (RUO) method, rely on the detection of the protein complement of the SARS-CoV-2. Corona viruses are unusually protein rich, which makes LC-MS combined with affinity enrichment an attractive approach to detect infection (https://elifesciences.org/articles/70843 ). The two most abundant proteins in SARS-CoV-2 are SPIKE (Spike glycoprotein) and NCAP (Nucleoprotein). SPIKE is responsible for the primary interaction and infection of the host. Changes within the glycan motif and mutation of the amino acid sequence are therefore of great concern from a virology perspective. NCAP is a structural component of the viral particle and involved in replication and transcription of the genome of the virus. It is also the most abundant protein of SARS-CoV-2 and detected by means of the SARS-CoV-2 LC-MS Kit (RUO) (. https://www.waters.com/waters/en_US/SARS-CoV-2-LC-MS-Kit-(RUO)/nav.htm?cid=135083368&locale=en_US) method; hence, it is also critical to monitor changes in the amino acid composition of NCAP for both new and circulating SARS-CoV-2 variants since they can affect affinity enrichment efficiency.
