Membranous Nephropathy

Many diseases can affect your kidney function by attacking and damaging the glomeruli, the tiny filtering units inside your kidney where blood is cleaned. Glomerular diseases include many conditions with many different genetic and environmental causes. Membranous nephropathy (MN) is a type of glomerular disease and is an autoimmune disease. An autoimmune disease is caused when your body’s defense system turns against you and harms your body when it should be protecting you. Your defense system is known as your immune system which is “turned on” by glomerular disease. MN is one of the most common causes of nephrotic syndrome. When your immune system attacks the glomeruli in membranous nephropathy, it causes changes to the filters that lead you to lose large amount of protein into the urine. If this continues at high levels, it can eventually lead to kidney failure.

Immunoglobulin A Nephropathy (IgAN )

IgAN is an abbreviation for Immunoglobulin A Nephropathy and is also known as Berger’s disease. It is a critical progressive, autoimmune condition that results in the glomeruli inside the kidneys becoming damaged and inflamed. After some time, the inflammation can adversely affect the kidneys’ ability to filter waste from the blood, leading to kidney disease if not correctly and promptly treated.

Immunoglobulin A nephropathy reduces the glomerular filtration rate (how the kidneys filter wastes from the blood) and causes large amounts of protein to enter the urine. The protein makes the urine appear foamy. This condition affects between 130,000–150,000 people in the United States, and its causes are still not completely understood.

Purpose Of Our Trial

It is our aim to help persons affected by IgAN to receive effective treatments, and as such, this trial is intended to evaluate the potential benefit of sparsentan on kidney function. We will take an in-depth look at how it affects protein in urine and changes the estimated glomerular filtration rate (eGFR) compared to the current standard treatment.

Benefit Of Our Trial

Unfortunately, up to 50% of persons diagnosed with IgAN progress to end-stage kidney disease within the period of ten to twenty years, and some in less. Treating this condition requires a lot of time and financial resources since dialysis and kidney transplants are necessary after some time.
These strenuous cost burdens, coupled with the fact that no cure currently exists for IgA nephropathy, are a few reasons why participating in trials such as these are beneficial. Understanding how sparsentan works against IgAN can save time, money, energy, and lives.

FSGS — Prolato 02/09/2021

FSGS

Focal segmental glomerulosclerosis (FSGS) is a condition affecting the glomeruli, which is the area within the kidney that removes waste matter from the blood. In FSGS, scar tissue develops on the glomeruli and may be a complication of several other conditions, such as obesity, diabetes, other kidney diseases, and sickle cell. FSGS ought not to be taken lightly, as it can cause kidney failure, an even more serious condition for which treatment is either dialysis or kidney transplant. There are three types of FSGS; primary, secondary, and genetic.



Did you know that approximately 40% of adults and 20% of children with nephrotic syndrome are affected by FSGS? In fact, within the United States alone, FSGS accounts for 5%-20% of all people affected by end-stage renal disease. So chances are you may know someone who, unfortunately, is affected by FSGS.


Benefit Of Our Trial
Treating FSGS is typically a lengthy and costly process involving numerous specialists. Our Phase 2 trial, which lasts for a period of 12 weeks, aims to provide another management option that will simplify the treatment process for patients and healthcare providers.


Purpose Of Our Trial
This step in our journey of helping FSGS patients is geared at evaluating the efficacy, safety, tolerability, and pharmacokinetics of PF-06730512. After receiving the medication, our adult volunteers will be assessed to determine how the drug moves through the body, its effectiveness, possible side effects, etc.



You Or Your Loved One Can Be A Part Of Our Trial
To become a volunteer in our trial, you must at least be an adult, 18 years or older, with a confirmed diagnosis of FSGS. To view the other requirements for your participation, click here. Please note that even though you may be diagnosed with FSGS, you may not be approved for this trial if you:

are diagnosed with collapsing FSGS.
have advanced chronic changes on renal biopsy as evidenced by greater than 50% tubulointerstitial fibrosis.
have a body mass index (BMI) greater than 45 kg/m2.



In so many cases, the treatment administered to FSGS and other conditions is inefficient; and it is with being a part of a trial that patients actually find management that makes a difference and sometimes is life-saving. Diabetic Nephropathy
In the United States, about 100 million Americans are diabetics. There’s always a relationship between diabetes and kidney diseases. How is this so? Due to diabetes, the small blood vessels in the body are injured, leaving the kidney unable to purify your kidney.
Simply put, your kidney is doing its job at a poor level, or not even doing it at all. You have a build-up of waste in your body, spilling the waste and damaging other organs.

Why Are We Doing This Trial
With the current study under GFB-887-201, we hope to assess the safety and tolerability of the drug not only for patients of diabetic nephropathy, but also with focal segment glomerulosclerosis, and treatment-resistant minimal change diseases.
Across the United States, we’ll be bringing on board 125 people for this trial. We’ll be randomizing the participants to receive 3 ascending doses or a placebo.

Who Is Eligible For This Trial
To know your eligibility status for this trial, please click on this link; https://www.prolatoresearch.com/gfb887201. However, we are excluding patients with other forms of kidney disease other than DN, FSGS, and TR-MCD. Other exclusions include;
Any patient with a body mass above 40kg
With a history of malignancy,
History of organ transplants or kidney grafts,
Any history of drug, alcohol, and chemical abuse,
Any DN patients with a renal disease requiring immunosuppressive therapy,
In FSGS/TR-MCD patients, Currently on calcineurin inhibitors or history of resistance to calcineurin inhibitors, or Known history of severe or chronic hepatobiliary disease.
